چکیده

surface. MCR is unique in that it apparently inhibits DNA synthesis while binding to the plasma membrane (12, 13). This mechanism of action requires that the binding of MCR to the cell surface freezes on modifies the moleof the mom bnane in mediating cellular metabolism. Kunimotoetal. (13) hypothesize that the interaction between MCR and tumor cells is analogous to the interactions between bacteniocins and bacterial cells. Since MCR may serve as a valuable tool for elucidating membrane function and because the drug is active against the experimental model tumors that have some, although restricted, predictive value for clinical chemotherapy, de velopment of a sensitive, specific, and rapid method for estimating MCR concentrations in biological materials was warranted. This report describes the development of a ma dioimmune assay for MCR and use of the assay, as well as purified antimacromomycin antibodies, in the detection of MCR in body fluids and on tumor cell surfaces.

Radioimmune assay and characteristics of antibodies to macromomycin (NSC 170105).

A radioimmune assay for the antitumor agent, macromomycin, using purified, radioiodine-labeled macromomycin and antisera raised in rabbits against a carbodiimide-catalyzed macromycin-Limulus polyphemus hemocyanin complex has been developed. Radiolabeled macromomycin was prepared by direct iodination of the polypeptide antibiotic with the use of iodine monochloride or solid-state lactoperoxidase...

Antitumor activity of macromomycin B (NSC 170105) against murine leukemias, melanoma, and lung carcinoma.

Mice bearing either of the two rapidly growing mouse leukemias, L1210 or P388, or the slow-growing B16 melanoma responded to i.p. injections of Macromomycin B (NSC 170105) with significant increases in life-span. The maximal increases in life-span obtained in these experiments were 37% for L1210, 68% for P388, and 120% for B16. In addition, there were 7 of 30 cures for varying doses of Macromom...

Bactericidal activity of macromomycin, a antitumor antibiotic.

Susceptibility testing by the broth dilution method showed that all the gram-positive but only some of the gram-negative bacteria tested were susceptible to the antitumor antibiotic, macromomycin (MCR; NSC 170105). The minimal inhibitory concentration for the susceptible organisms was less than 3 mug/ml. The action of MCR was bactericidal; however, at very high concentrations (50 mug/ml and abo...

Effects of zinc ion on the conformation of antigenic determinants on insulin.

Comparison of c.d. spectra of Zn-insulin with Zn2+-free insulin demonstrated significant differences. It has been proposed that these differences are due to either changes in the structure of insulin monomers within aggregated insulins or the results of insulin aggregation. The effect of Zn2+ on the immunological activity of insulin indicated that the antigenic determinants of insulin were also...

Production of a macromomycin (MCR)-monoclonal antibody conjugate and its biological activity.

Macromomycin (MCR), an unique membrane-reactive anticancer antibiotic, was incubated with murine monoclonal anti-HLA IgG1 antibody (H-1) in the presence of carbodiimide. The resulting mixture was fractionated with a Sephadex G-200 column. The first and second fractions were shown to contain MCR-(H-1) conjugate by the elution profile, as well as by the Sarcina lutea growth inhibition assay and O...
